This comes after the prime minister being abandoned by newly-appointed ministers and more than 50 others in a rebellion that had left government dangerously close to paralysis.
Eight ministers, including two secretaries of state, have so far resigned in the last two hours, leaving Johnson isolated and powerless, a trend that has made the prime minister to bow to pressure, media reports say.
"Boris Johnson will resign as Conservative Party leader today," the BBC's political editor Chris Mason said.
After days of battling for his job, Johnson had been abandoned by all but a handful of allies.
This state of affairs is a far cry from when Johnson, 58, rose to power in 2019 with a large majority, capturing votes in parts of Britain that had never supported his Conservative Party before.
Even his finance minister, Nadhim Zahawi, who was only appointed to his post on Wednesday, had called on his boss to resign.
"This is not sustainable and it will only get worse: for you, for the Conservative Party and most importantly of all the country," he said on Twitter. "You must do the right thing and go now."
UK education minister Michelle Donelan, only promoted to the role two days ago amid a government exodus, quit on Thursday saying prime minister Boris Johnson had put her in "an impossible situation".
"I am deeply saddened that it has come to this, but as someone who values integrity above all else, I have no choice," she wrote in a statement posted on Twitter.
Johnson came under fire last year after it was discovered that he was throwing parties while the country was locked down with no one allowed to party.
